Gene expression is regulated at not only initiation but also many other stages that follow the initiation stage. The dynamic nature of transcription and its regulation, however, cannot be fully captured in the experiments at the ensemble level, and as a result several key questions concerning gene regulation mechanisms during transcription remain poorly answered. In this talk, I will introduce the single-molecule fluorescence assays that we developed, and discuss the mechanisms of the factor-independent and factor-dependent transcription terminations of E. coli Revealed by the utilization of the techniques.
